AB - The multigene <ce>family</ce> <nwt>of</nwt> <bt>cytosolic</bt> <protein>glutathione S-transferases</protein> (GSTs) consists <nwt>of</nwt> <ce>four</ce> classes (Alpha, Mu, <nwt>Pi</nwt>, and Theta), <ce>all</ce> <ce>involved</ce> in the detoxication <nwt>of</nwt> <ce>reactive</ce> electrophiles. The <bt>human</bt> Mu <ce>class</ce> GSTs consist <nwt>of</nwt> at least <ce>four</ce> <ce>expressed</ce> <mcg>isozyme</mcg> <mcg>subunits</mcg>, <nwt>GST</nwt> <protein>M1</protein>, <nwt>GST</nwt> <protein>M2</protein>, <nwt>GST</nwt> <protein>M3</protein>, and <nwt>GST</nwt> <protein>M4</protein>, which have 70-90% <ct>amino</ct> <ct>acid</ct> <mcg>sequence</mcg> identity. The <mcg>gene</mcg> and <nwt>cDNA</nwt> <mcg>sequences</mcg> for <nwt>GST</nwt> <protein>M4</protein> have been determined recently (K. E. Comstock, K. J. Johnson, D. Rifenbery, and W. D. Henner, J. Biol. Chem. (1993) 268, 16958-16965). Cloning <nwt>of</nwt> <nwt>GST</nwt> <protein>M4</protein> <nwt>cDNA</nwt> into an Escherichia <bt>coli</bt> <ce>expression</ce> <ce>system</ce> permitted the <ce>production</ce> <nwt>of</nwt> the corresponding <mcg>protein</mcg>. The <mcg>enzyme</mcg> <nwt>was</nwt> purified and shown to have a relatively <ce>low</ce> <ce>specific</ce> <ce>activity</ce> with the standard <nwt>GST</nwt> <ct>substrate</ct> 1-chloro-2,4-dinitrobenzene (1.4 +/- 0.2 mumol min-1 <nwt>mg</nwt>-1 <mcg>protein</mcg>), but an <ce>activity</ce> equivalent to <ce>other</ce> Mu <ce>class</ce> enzymes with <ce>other</ce> tested <ct>substrates</ct>. The <mcg>protein</mcg> <ce>forms</ce> <ce>functional</ce> dimers composed <nwt>of</nwt> <mcg>subunits</mcg> with a M(r) <nwt>of</nwt> approximately 26,400. A detailed comparison <nwt>of</nwt> the <ce>activity</ce> with various <ct>substrates</ct> and inhibitors <nwt>was</nwt> performed <ce>between</ce> <nwt>GST</nwt> <protein>M4</protein>-4 and <ce>other</ce> <bt>human</bt> Mu <ce>class</ce> GSTs, <nwt>GST</nwt> M1a-1a, <nwt>GST</nwt> <protein>M2</protein>-2, and <nwt>GST</nwt> <protein>M3</protein>-3, produced in <bt>bacterial</bt> <ce>expression</ce> systems. Despite the <ce>high</ce> <ce>level</ce> <nwt>of</nwt> <ct>amino</ct> <ct>acid</ct> <mcg>sequence</mcg> identity, the enzymatic properties <nwt>of</nwt> these enzymes were quite different. Comparisons with the crystallographic <ce>structure</ce> <nwt>of</nwt> a <ce>homologous</ce> <nwt>rat</nwt> <nwt>GST</nwt>, <nwt>GST</nwt> 3-3, indicate that a <ce>number</ce> <nwt>of</nwt> the nonconserved <ct>amino</ct> <ct>acid</ct> <bt>residues</bt> can be assigned to the <ce>putative</ce> <ce>active</ce> <ce>site</ce> <nwt>of</nwt> <nwt>GST</nwt> <protein>M4</protein>-4. This suggests that diversification in the evolution <nwt>of</nwt> these <bt>genes</bt> <nwt>has</nwt> occurred primarily in the <ct>substrate</ct> <ce>binding</ce> regions to cope with an increasing variety <nwt>of</nwt> foreign compounds. <ce>University</ce>, Portland 97201.
